A recent study published in the British Medical Journal links the Depo-Provera injection, which contains medroxyprogesterone acetate, to an increased risk of developing intracranial meningiomas, particularly with prolonged use (≥1 year). The French study from March 2024 found that high-dose medroxyprogesterone injections (150 mg) were associated with a heightened risk of meningiomas requiring surgery. The study included 108,366 women, with 18,061 of those women having had brain surgery to remove a meningioma.
What is Depo-Provera?
Depo-Provera (also known as medroxyprogesterone acetate or the depo shot) is primarily used as a contraceptive to prevent pregnancy. It contains medroxyprogesterone acetate, a synthetic form of the hormone progesterone, and is administered as an intramuscular shot every three months. Depo-Provera works by preventing ovulation, thickening cervical mucus to block sperm, and thinning the uterine lining to prevent implantation. In addition to birth control, it can also be used to manage conditions like endometriosis, abnormal uterine bleeding, and as part of hormone therapy.
Depo-SubQ Provera 104 is administered as a subcutaneous injection, which means it is injected just under the skin, typically in the abdomen or thigh. The injection is usually given every 12 to 14 weeks, and it is done using a small needle to ensure the medication is delivered into the fatty tissue.

Most meningiomas are benign and slow-growing, but they can still cause issues by compressing nearby brain tissue.
